Matrixyl
A lipopeptide that stimulates collagen production by mimicking the body's natural collagen-building signals, one of the most proven and widely used anti-aging cosmetic peptides.
Overview
Matrixyl (Palmitoyl Pentapeptide-4, or pal-KTTKS) is a synthetic lipopeptide developed by Sederma (now part of Croda International) that has become one of the most established and evidence-supported anti-aging cosmetic peptides. It consists of a pentapeptide sequence (KTTKS) derived from the C-terminal propeptide of type I procollagen, linked to a palmitoyl (palmitic acid) lipid tail that enhances skin penetration.
The mechanism of Matrixyl differs fundamentally from neuromuscular peptides like Argireline and Snap-8. Rather than reducing muscle contraction, Matrixyl works by stimulating the production of extracellular matrix components — particularly collagen, elastin, and fibronectin — by fibroblasts in the dermis. The KTTKS sequence acts as a matrikine, a messenger peptide released during normal collagen turnover that signals fibroblasts to produce new collagen. By providing this signal exogenously, Matrixyl essentially tricks the skin into boosting its own collagen production.
Clinical evidence for Matrixyl is among the strongest in the cosmetic peptide category. A pivotal double-blind, placebo-controlled study showed that 12 weeks of treatment with Matrixyl reduced wrinkle depth by up to 68% in some parameters, with effects comparable to retinol but with significantly less skin irritation. Additional studies have demonstrated improvements in skin firmness, thickness, and smoothness.
Matrixyl has been expanded into several variants. Matrixyl 3000 combines Palmitoyl Tripeptide-1 and Palmitoyl Tetrapeptide-7, targeting both collagen production and inflammation. Matrixyl Synthe'6 (Palmitoyl Tripeptide-38) targets six major components of the skin matrix. These variations allow formulators to target different aspects of skin aging. The Matrixyl family of peptides has become a gold standard in cosmetic anti-aging and is included in products from virtually every major skincare brand.
